The 2-side refers to the spacing on the court where the 45 and corner are filled. The 2-side break is a fast break where these spots are filled. It is a transition offense principle designed to generate high-quality shots early in the shot clock.
After gaining possession either on a defensive rebound or inbounding the ball after a score, the ball handler picks a side of the floor to bring the ball up the court. The off-ball players are sprinting to the weakside of the court to create a 2-side.
This takes advantage that teams often load to the basket and then the ball in transition defense. This means that if there are 2 players back to guard the 2 players on the weakside, one of those players will be protecting the basket. See video below.
